In the following three articles about chitosan in connection with curcumin will be introduced. Curcumin is the colorant and the effective ingredient of the medical plant curcuma. Curcumin can be used in different medical fields, e. g. Alzheimer’s disease or cancer diseases. It can also be used as prophylaxis of lung diseases, intestine diseases and liver diseases.

Elaboration of chitosan-coated nanoparticles loaded with curcumin for mucoadhesive applications.

The development of chitosan coated polycaprolactone (PLC) nanoparticles containing curcumin for buccal delivery was the goal of this study. PLC is a biodegradable synthetic material. Preparations with controlled drug release are one possibility of the range of PLC  applications in the field of medicine. Curcumin needs such a carrier to reach the targeted application location in desired concentration. The particles were prepared via nano precipitation method. To optimize the preparation conditions different molar masses and concentrations of chitosan and different concentrations of triblock surfactant poloxamer (PEO-PPO-PEO) were used.

Chitosan coated particles showed a positive surface charge. The mean particle radius was between 144 and 125 nm. Dynamic light scattering (DLS) has shown that the particles are monodisperse because the polydispersity indices were lower than 0.3. The nanoparticle tracking analysis (NTA) was well accorded to the results of the DLS. The mean drug content of the colloidal systems was about 460µg/ml and the encapsulation efficiency was higher than 99%. The well mucin interacting particles coated with chitosan illustrated their suitability for mucoadhesive applications.

Oral delivery of curcumin bound to chitosan nanoparticles cured Plasmodium yoelii infected mice.

Plasmodia are important in the field of malaria research. They serve as model organisms and can be used in laboratory to infect mice and rats in part. The study especially deals with Plasmodium yoelii (P. yoelii) infected mice healing.

Curcumin is attributed to have an anti malarial activity. Because of its poor bioavailability and chemical instability, it depends on a carrier to achieve the desired effect. In this study curcumin was bound to chitosan nanoparticles to enhance the mentioned properties.

The research showed the following results:
The degradation of curcumin bound to chitosan was less rapidly in comparison to free curcumin. Mice RBCs (red blood cells) assimilated the bound curcumin much better than the non-bound curcumin. Oral delivered curcumin loaded chitosan nanoparticles enhanced the bioavailability of curcumin in plasma and RBCs. While P. yoelii infected mice died 8 to 9 days post infection, nanoparticle (without curcumin) fed mice survived for 5 more days. Oral delivered free curcumin (1mg per day for seven days) kept a third of the infected mice alive. All mice survived by oral delivered curcumin bound to chitosan nanoparticles (same dosage).

The scientists assume that the bound curcumin was also enhanced regarding the chemical stability.

Quantitative Determination of Proteins Based on Strong Fluorescence Enhancement in Curcumin-Chitosan-Proteins System.

Among other things fluorescence is used in medicine to localize substances respectively via fluorescence it is possible to make biological procedures viewable.

The study showed that the fluorescence intensity of curcumin can clearly be enhanced by albumins in the presence of chitosan. Albumins are so-called globular proteins. In this study the following albumins have been used:

  •     BSA (bovine serum albumin) and
  •     HAS (human serum albumin)

The fluorescence increase is the basis of a new fluorimetric method to determine protein concentration.

Synergetic effects of favourable hydrophobic microenvironment provided by BSA and chitosan as well as efficient intermolecular energy transfer between BSA and curcumin are the reason for the increased fluorescence of curcumin. Curcumin is able to bind itself to the BSA chitosan complex. Chitosan plays a key role because by shortening the distance between BSA and curcumin chitosan promotes the energy transfer process.

Im Folgenden werden drei Artikel zum Thema Chitosan im Zusammenhang mit Kurkumin vorgestellt. Kurkumin ist Farbstoff und wirksamer Bestandteil der Heilpflanze Kurkuma. Kurkumin wird in verschiedensten medizinischen Bereichen verwendet. So z. B. bei Alzheimer- oder Krebserkrankungen. Auch zur Vorbeugung von Lungen-, Darm und Lebererkrankungen wird Kurkumin eingesetzt.

Ziel der Untersuchung war die Herstellung eines mit Chitosan beschichteten Polycaprolacton (PLC) Nanopartikels, der Kurkumin enthält und zur bukkalen Verabreichung (über die Wange) eingesetzt werden kann. PLC ist ein biologisch abbaubarer Kunststoff, der in der Medizin u. a. für Präparate mit kontrollierter Wirkstofffreigabe verwendet wird. Kurkumin benötigt ein solches Transport-Vehikel, um den Anwendungsort in gewünschter Konzentration zu erreichen.

Die Präparierung dieser Partikel wurde mittels Nanopräzipitation vorgenommen. Um die Herstellungsbedingungen zu optimieren, wurden verschiedene Molmassen und Konzentrationen von Chitosan sowie verschiedene Konzentrationen eines 3er Blocks aus Tensid Poloxameren (PEO-PPO-PEO) verwendet.

Chitosan beschichtete Partikel wiesen eine positive Oberflächenladung und eine mittlere Partikelgröße mit einem Radius von 114 und 125 nm auf. Die Analyse mittels Lichtstreuung (DLS: Dynamic light scattering) konnte zeigen, dass die Partikel monodispers sind, da die Polydispersitätsindizes unter 0.3 lagen. Die Nanopartikel-Tracking-Analyse (NTA) stimmte in den Ergebnissen gut mit denen der DLS überein.

Die kolloidalen Systeme wiesen einen mittleren Wirkstoffgehalt von 460µg/ml und eine Verkapselungseffizienz von über 99% auf. Die gute Interaktion der Chitosan beschichteten Partikel mit der Schleimhaut, verdeutlicht die Eignung für mucoadhäsive Anwendungen.

Plasmodien spielen eine wichtige Rolle im Bereich der Malariaforschung. Sie dienen als Modellorganismen, mit denen im Labor Mäuse, teilweise auch Ratten, infiziert werden können. Die Studie befasste sich speziell mit der Heilung Plasmodium yoelii (P. yoelii) infizierter Mäuse.

Dem Kurkumin werden Malaria kurierende Eigenschaften zugeschrieben. Aufgrund seiner geringen biologischen Verfügbarkeit und chemischen Instabilität ist es allerdings auf ein Träger-Vehikel angewiesen, um die gewünschte Wirkung zu erzielen. In dieser Studie wurde Kurkumin an Chitosan-Nanopartikel gebunden, um die genannten Eigenschaften zu verbessern.

Die Untersuchung zeigte folgende Resultate:

  • Im Vergleich zu ungebundenem Kurkumin, zeigte sich bei Chitosan-Nanopartikel gebundenem Kurkumin ein weniger schneller Abbau.
  • Die roten Blutkörperchen (RBCs: red blood cells) der Mäuse nahmen zudem das gebundene Kurkumin wesentlich besser auf als das ungebundene.
  • Kurkumin beladene Chitosan-Nanopartikel verbesserten bei oraler Verabreichung die biologische Verfügbarkeit des Kurkumins im Plasma und den RBCs.
  • Während P. yoelii infizierte Mäuse nach 8 bis 9 Tagen (nach der Infektion) starben, lebten jene Mäuse, die mit einfachen Chitosan-Nanopartikeln gefüttert wurden, weitere 5 Tage.
  • Bei einer oralen Verabreichung ungebundenen Kurkumins (1mg täglich über 7 Tage) überlebte 1/3 der infizierten Mäuse.
  • Bei einer oralen Verabreichung von Kurkumin-Chitosan-Nanopartikeln (gleiche Dosis) überlebten alle Mäuse.

Die Forscher gehen davon aus, dass das gebundene Kurkumin auch in seiner chemischen Stabilität verbessert wurde.

In der Medizin dient die Fluoreszenz u. a. zur Lokalisierung von Substanzen bzw. können mit Hilfe. von Fluoreszenz auch biologische Vorgänge sichtbar gemacht werden.

Die vorliegende Studie zeigt auf, dass die Fluoreszenz-Intensität von Kurkumin deutlich durch Albumine in Verbindung von Chitosan verstärkt werden kann. Albumine sind so genannte globuläre Proteine. In der Studie verwandte Albumine waren:

  • Rinderalbumin (BSA: bovine serum albumin) und
  • Humanalbumin (HSA: human serum albumin).

Die Verstärkung der Fluoreszenz bildet die Grundlage einer neuen fluorimetrischen Methode zur Bestimmung der Protein-Konzentration.

Die erhöhte Fluoreszenz von Kurkumin resultiert aus einem Synergieeffekt der günstigen hydrophoben Mikroumgebung von BSA und Chitosan und dem effizienten intermolekularem Energietransfer zwischen BSA und Kurkumin. Kurkumin ist in der Lage, sich mit dem BSA-Chitosan-Komplex zu verbinden. Chitosan spielt dabei eine Schlüsselrolle und begünstigt den Prozess des Energietransfers, indem es den Abstand zwischen BSA und Kurkumin verringert.
